NBA 2K10 comes with a new mode called My Player. Unfortunately 2K Sports forgot to add some settings to change match time and difficulty. Many started to complain already so until the first official patch you can use this guide to change things.
If you want to edit foul sliders, gameplay sliders, etc., look at Memory editing sliders (Cht file included) - 2K Sports Forums
To edit "Quarter Length" in "Minutes" : 5443ABC
To edit "Simulated Quarter Length" in "Minutes" : 5443AC0
To edit "Difficulty" : 54439C4
For NBA2K10 on the PC, you can use a memory editor to adjust these values where editing it in the options is not available.
1.) Download and install a program called tsearch. This is the program that will be used to edit the memory.
2.) Run NBA 2K10 and load your "My Player" save file. Hold your "Alt" key and press "Tab" to minimize the game.
3.) Run "tsearch.exe".
4.) Click the "Open Process" button in the top left. In the menu that comes up, chose "nba2k10.exe".
5.) Click "Add New Cheat", which is an icon of a sheet of paper with a folded top right corner under the "Cheats List" icon. It's part of the right window in the middle of the program.
6.) Set the description to whatever you'd like. I set mine to Quarter Length. In the "Address" box, put: 5443ABC. Set the "Type" to "4 Byte". Then click the "Set" button to set a hotkey.
7.) Click "Click Here To Set" and set a key. I set my key to "F8". Then set the "Action" to "Set Value". Then change tha "Value" from 0 to the amount of minutes you want the quarters to be. 8 seems to be the most realistic for in-game quarter length, while 12 minutes is the best for simulated quarter length (when you're not playing the game at all, just using supersim from the menu for association mode). I personally set my value to "8".
8.) Once this is done, click the "NBA 2K10" button in your task bar at the bottom of your screen to bring the game back up. Once it's up, and you should be at the "My Player" "Home" screen, you can press F8 or whatever key you set it to. Then, the next time you play a game, it should have 8 minute quarters.
9.) Alternately, from step 6, instead of editing quarter length, to change difficulty:
In the "Address" box, put: 54439C4. Set the "Type" to "4 Byte". Then click the "Set" button to set a hotkey.
10.) Click "Click Here To Set" and set a key. I set my key to "F9". Then set the "Action" to "Set Value". 0-Rookie, 1-Pro, 2-All Star, 3-Superstar, 4-Hall of Fame. So if you set the value to 4 and load the game back up, pressing F9 will set each game's difficulty to Hall of Fame that you play from that point on, starting at the next game.
*When you are ready don't close tsearch. Restore NBA 2K10 window play at least one game, save the game and the changes will be also saved.
